Lady Mary was the only daughter and third child of Francis Osborne, 5th Duke of Leeds KG, PC (1751-1799) by his first wife, Lady Amelia Darcy (1754-1784). In 1801, Lady Mary married the Whig politician, Thomas Pelham, 2nd Earl of Chichester PC, PC (Ire), FRS (1756-1826). John Thomas (1811-1894), Lady Catherine Georgiana (1814-1885) and Lady Lucy Anne (1815-1901). Their second son, Frederick, became a Rear-Admiral in the Royal Navy and their third son, John, became Bishop of Norwich. Their eldest daughter, Amelia married Major General Sir Joshua Jebb, the Surveyor General of Prisons and designer of Pentonville Prison on 5 September 1854.
